Israel eases building materials blockade on Gaza

JERUSALEM (AP) ? Israel has started allowing long-banned building materials into the Gaza Strip, its first key concession to the Palestinian territory's ruling Hamas movement under the cease-fire that ended eight days of fighting last month, the military said Monday.

The military says the shipments will continue so long as the border remains quiet. But a Hamas official said the amount sent so far is "cosmetic," and Gaza economists say it would take years of round-the-clock shipments to even make a dent in the gap left by five years of blockade.

Israel imposed a wide-ranging embargo on Gaza after Hamas seized it in 2007. Building materials like cement, gravel and metal rods were largely banned, as Israel argued they could be used to make fortifications and weapons to attack the Jewish state.

The military says it began allowing shipments of gravel to Gaza's private sector on Sunday because the Israeli attacks on Hamas' military operations in November cowed the militant group into quiet.

After the November hostilities, Israel and Hamas began indirect, Egyptian-led talks over new border arrangements. The militant group wants Israel to lift what remains of a sweeping land and naval embargo it imposed after the Hamas takeover. In return, Israel demands an end to arms smuggling into Gaza.

With limited exceptions, Israel had blocked construction materials from entering Gaza.

"Now we're talking about a permanent easing," said Maj. Guy Inbar, a military spokesman, adding that 20 truckloads a day could enter Gaza, depending on Palestinian demand. Other concessions could follow, he said.

"The longer the calm persists, the more we'll weigh additional easings of restrictions that will benefit the private sector," he said.

Israel recently authorized the entry of 60 trucks and buses for the first time since Hamas' 2007 Gaza takeover, though there are conflicting reports on whether vehicles have actually gone through.

Another major concession the Gazans seek would be a lifting of a near-ban on exports from the impoverished territory. Exports, especially to the West Bank, the Palestinian territory on the opposite side of Israel, once formed the backbone of Gaza's economy.

Exports might be expanded, Inbar said, "depending on the continuation of the calm."

Critics contend the export ban punishes ordinary Gazans instead of pressuring Hamas, hurting four in five Gaza factories and contributing heavily to an unemployment rate of about one-third of the work force. Eighty percent of Gaza's 1.6 million people rely on U.N. handouts.

Israel lifted its restrictions on consumer goods entering Gaza overland after a deadly Israeli naval raid on a blockade-busting flotilla in 2010 drew international attention to the Israeli embargo. But the blockade on construction materials remained intact, save for shipments used to build U.N. schools and a pilot project of shipments to the private sector a year ago.

"The Israelis promised to undertake further measures to alleviate the difficult economic situation in Gaza as a result of the calm," said Palestinian crossing official Raed Fattouh in Gaza, confirming that the Israelis had agreed to send in 20 trucks of gravel daily, five days a week. "This move had been expected as part of the deal."

Israel has not eased its naval blockade of the territory, which it says is imperative to keep weapons from being smuggled into Gaza by sea.

Egypt, which had joined the Israeli blockade, similarly eased its own restrictions on Saturday, allowing in 1,400 tons of gravel paid for by Qatar. The oil-rich emirate recently pledged $425 million to build housing, schools, a hospital and roads in Gaza as part of its attempt to build its influence in Palestinian politics and its power in the region, at the expense of regional rival Iran, Hamas' longtime patron.

Shipments from Egypt are expected to be ramped up to 4,000 tons daily, said Yassir al Shanti, Gaza's deputy minister of housing and public works. He estimated Gaza needs up to 3 million tons of gravel to build roads and that the Qatar-funded projects need more than 1 million tons.

The shipments from Egypt were launched following consultation with Israeli officials, who were in Cairo Thursday to discuss the cease-fire and other matters, an Egyptian official said last week.

Under former President Hosni Mubarak, Israel's longtime ally, Egypt had poor relations with Hamas, and teamed up with Israel to blockade Gaza. Egypt's new president, Mohammed Morsi, comes from Hamas' parent group, the Muslim Brotherhood, and has vowed not to abandon the Palestinians. But he is moving cautiously, in part to avoid alienating Cairo's biggest patron, the United States.

Palestinian economist Mouin Rajab said the new shipments would go only a small way to meet the needs Gaza has accumulated throughout six years of blockade, during which time Hamas and Israel warred twice.

"Gaza needs more than what Israel has allowed and what Egypt has promised to allow. We are talking about six years of blockade, no real economy and no projects in addition to what Gaza lost during two wars in 2009 and 2012."

"This amount which has been sent by the Israelis still is cosmetic," a Hamas government official in Gaza said. "Israel, according to the understanding, should allow more building materials into Gaza as part of the understandings reached by Cairo. We are waiting and we told the Egyptians that."

He spoke on condition of anonymity because he was not authorized to discuss the understandings.

Reconstruction since the 2009 fighting has been slow, in large part because of the blockades. To make up the shortage, a bustling smuggling industry through underground tunnels along the Egyptian border has sprung up. While prices for key construction goods have come down, they still remain expensive for the majority of Gaza's 1.6 million people, 80 percent of whom rely on U.N. handouts.

Israel and Hamas shun each other, so Egypt is mediating the new border arrangements. A Hamas official, speaking on condition of anonymity because he wasn't authorized to disclose confidential contacts, said a Hamas delegation arrived Sunday night in Cairo to meet with Egyptian security officials for a second round of talks on the border arrangements.

Teenagers without Internet access at home are educationally disadvantaged, UK study suggests

Dec. 30, 2012 ? A major in-depth study examining how teenagers in the UK are using the internet and other mobile devices says the benefits of using such technologies far outweigh any perceived risks.

The findings are based on a large-scale study of more than 1,000 randomly selected households in the UK, coupled with regular face-to-face interviews with more than 200 teenagers and their families between 2008 and 2011.

While the study reflects a high level of parental anxiety about the potential of social networking sites to distract their offspring, and shows that some parents despair at their children's tendency to multitask on mobile devices, the research by Oxford University's Department of Education concludes that there are substantial educational advantages in teenagers being able to access the internet at home.

Teenagers who do not have access to the internet in their home have a strong sense of being 'educationally disadvantaged', warns the study. At the time of the study, the researchers estimated that around 10 per cent of the teenagers were without online connectivity at home, with most of this group living in poorer households. While recent figures from the Office of National Statistics suggest this dropped to five per cent in 2012, the researchers say that still leaves around 300,000 children without internet access in their homes.

The researchers' interviews with teenagers reveal that they felt shut out of their peer group socially and also disadvantaged in their studies as so much of the college or school work set for them to do at home required online research or preparation. One teenager, whose parents had separated, explained that he would ring his father who had internet access and any requested materials were then mailed to him through the post.

Researcher Dr Rebecca Eynon commented: 'While it's difficult to state a precise figure for teenagers without access to the internet at home, the fact remains that in the UK, there is something like 300,000 young people who do not -- and that's a significant number. Behind the statistics, our qualitative research shows that these disconnected young people are clearly missing out both educationally and socially.'

In an interview with a researcher, one 14-year old boy said: 'We get coursework now in Year 9 to see what groups we're going to go in Year 10. And people with internet, they can get higher marks because they can like research on the internet?my friends are probably on it [MSN] all the day every day. And like they talk about it in school, what happened on MSN.'

Another teenager, aged 15, commented: 'It was bell gone and I have a lot of things that I could write and I was angry that I haven't got a computer because I might finish it at home when I've got lots of time to do it. But because when I'm at school I need to do it very fast.'

Strikingly, this study contradicts claims that others have made about the potential risks of such technologies adversely affecting the ability of teenagers to concentrate on serious study. The researchers, Dr Chris Davies and Dr Rebecca Eynon, found no evidence to support this claim. Furthermore, their study concludes that the internet has opened up far more opportunities for young people to do their learning at home.

Dr Davies said: 'Parental anxiety about how teenagers might use the very technologies that they have bought their own children at considerable expense is leading some to discourage their children from becoming confident users. The evidence, based on the survey and hundreds of interviews, shows that parents have tended to focus on the negative side -- especially the distracting effects of social networking sites -- without always seeing the positive use that their children often make of being online.'

Teenagers' experiences of the social networking site Facebook appear to be mixed, says the study. Although some regarded Facebook as an integral part of their social life, others were concerned about the number of arguments that had escalated due to others wading in as a result of comments and photographs being posted.

The age of teenagers using Facebook for the first time was found to go down over the three year period from around 16 years old in 2008 to 12 or 13 years old by 2011. Interviews reveal that even the very youngest teenagers who were not particularly interested felt under some peer pressure to join. But the study also suggests that the popularity of Facebook is waning, with teenagers now exploring other forms of social networking.

Dr Davies commented: 'There is no steady state of teenage technology use -- fashions and trends are constantly shifting, and things change very rapidly when they do change.'

The research was part funded by Becta, the British Educational Communications and Technology Agency, a non-departmental public body formed under the last Labour government. The study findings are contained in a new book entitled, Teenagers and Technology, due to be published by Routledge in January 2013.

366 Days, 366 Books

Book a Day

Like many of you, I have a nagging problem: I?m not a heroin user. Nor am I a knuckle-cracker, a nail-biter, or a thumb-sucker. I (usually) pay off my credit cards every month. I travel, I see my family, I?m not stressed. And the big three?drinking, dieting, and smoking cessation? I?m good.

This is all great for my insurance rates, but not when trying to determine a New Year?s resolution. So what can someone like me do for 2013? Other than tweet a humblebrag on the subject? (I?m no good at New Year?s resolutions ? life is too perfect I guess LOL!)

I had the same dilemma last year, and my solution, while ingenious, turned out to be quite a challenge. I decided that I would read more. Not a book a month, or even a book a week. One entire book every day. Three hundred sixty-six books by year?s end. ?Challenge accepted!? the Barney Stinson of my soul boasted. No sweat.

If you?re like me, the list of books you want to read is a hydra. Cutting one head off (that is, reading a book) just scribbles new titles onto the list. I?m never ahead. My whole life I?ve been fighting this hydra tactically, trying to bleed a head dry by reading, say, one of Bob Woodward?s or Neal Stephenson?s books every year. But they keep writing more books! Time for a new solution: overwhelming firepower. Force-feed my eyeballs like every tomorrow was a final exam. By year?s end, my 366 books would have that hydra on its knees.

And, believe it or not, it worked.

In the cold light of New Year?s morning, my challenge seemed a bit daunting. So I gave myself a week. If I didn?t have seven books finished by Jan. 7, I obviously wasn?t going to be able to keep such a meth-addled pace.

The resolution almost didn?t last 24 hours, but New Year?s night I forced my eyelids open and read a short horror novel. The following day I finished Cinderella Ate My Daughter, which, despite its title, was not a horror novel but a pop-culture history I?d been working on for a few days. The day after that I peeled off a Walking Dead collection. Driving to work, I hit the final disc of a Nelson DeMille audiobook. The night after that, I read the final chapter of a Narnia book to my daughter at bedtime.

Seven days, seven books. So far, so good. A good six of those books were timing and luck?it can take me three weeks to finish an audiobook, and more than a month to finish a bedtime story. But I gave it another week. And another. Soon my every waking thought was on reading.

The prime directive for this sort of project, you might think, is to pick short books. I don?t deny that 2012 was not the year for me to launch into Terry Goodkind. Want some Tolstoy? The Forged Coupon, not War and Peace. But as it turned out, Read Short Books was rule No. 2.

No, my prime directive was: no min-maxing. In Dungeons and Dragons, ?min-maxing? is focusing on one character attribute to the exclusion of everything else. (Sheldon Cooper from The Big Bang Theory is an excellent example of someone who min-maxes intelligence, stealing points from charisma and dexterity.) If you?ve ever been in a team sport, school play, or med school, you know how that commitment supersedes all others. Parties are missed, sleep is skipped, emails go unreturned. ?I can?t?I have practice/rehearsal/a corpse to dissect.?

U.S. Education Dept. Offers Tools for Evaluating Ed. Tech. - Digital ...

Education technology, to state the obvious, is everywhere. But how can school officials judge the effectiveness of the myriad tools and products being marketed to them, and their usefulness in terms of meeting the particular needs of teachers and students?

The U.S. Department of Education's Office of Educational Technology has released a draft report, "Expanding Evidence Approaches for Learning in a Digital World," designed to offer the education community some guidance for navigating the crowded tech landscape.

The report is meant to provide approaches for school officials and others seeking to gather evidence on digital learning systems, guidance that can be adapted to the needs of individual schools and districts. The document draws from the perspectives of education researchers, school technology developers, and educators themselves.

The report was released earlier this month in draft form for public comment, Karen Cator, the director of the office of educational technology, explained in an e-mail. Cator also discussed the goals of the document in a recent online blog post.

The document includes a "framework" meant to help educators and others evaluate the uses of education technology. One of the goals of the framework is to give school officials "greater confidence that investments in cost-effective and cost-saving technology-based interventions are wise, capable producing the outcomes sought," the report says.

That framework includes an evidence "reference guide," which focuses primarily on six approaches for using evidence to evaluate school technology, as well as other approaches commmonly used in education. It presents readers with an evidence "decision-making model," that can be used to gather evidence on digital learning resources, once they have been selected, so that they can be implemented effectively.

And it offers scenarios describing how various evidence standards might be used in hypothetical situations?for a superintendent trying to choose a technology-based math curriculum aligned with the Common Core State Standards, for instance, or a mother attempting to research the most appropriate online games, apps, and websites to help her soon improve his vocabulary.

"The need for expanded approaches to evidence that take advantage of and solve challenges created by digital learning resources is not new," the authors say. "What is new is the increased number and sophistication of digital learning resources and the vast amounts of data those systems generate while in use."

Schools are operating amid a "rapid rate of consumer adoption of these resources," the report continues. "These developments provide the opportunity to ask and answer these essential questions: What is appropriate evidence under which circumstances? How do we obtain it? How do we use it?"

The approaches for using evidence described in the report are also meant to address five overriding challenges in digital education, the authors say: ensuring that tech resources promote deep learning; creating "adaptive learning systems" that allow for personalized approaches for different students; combining and analyzing data from different sources to provide more targeted help for students; improving assessment through technology; and helping educators make informed choices about digital resources.

The Diver's Skin Is Kept Completely Dry And The Material Of The

The diver?s skin is kept completely dry and the material of ? for obvious reasons ? that many find themselves addicted to after just the first time. If you started with the basics with scuba diving flippers, you have an enjoyable ? and memorable ? scuba experience. But the art of scuba diving has opened up the possibilities swimming capabilities, be very uncomfortable and even dangerous while taking a dive. As you gaze at the shelf of scuba diving flippers, you will be amazed to diving flippers in the market, choosing a pair can be very confusing.

The Buoyancy Compensator, such as such as a back-mounted wing push needed for scuba diving and are usually used for snorkeling. Before taking up scuba diving as a recreational sport, it is from the water, and should be no lesser than 30 cms in length. It is not important how long an instructor has been training waste a lot of money if decides to stop doing this activity. Recently, some companies have created paddle fins that have pivoting blades that can angle to that allow underwater verbal communication, diving helmets using surface supplied diving equipment etc. There are however divers who dive for commercial purposes like for the purpose equipment ? is placed over the mouth, nose, and eyes of the diver.

The factor that could determine future breast cancer treatment

Dec. 27, 2012 ? Australian scientists have shown how a 'transcription factor' causes breast cancer to develop an aggressive subtype that lacks sensitivity to estrogen and does not respond to anti-estrogen therapies such as Tamoxifen and aromatase inhibitors.

Transcription factors are molecules that switch genes on or off. In this case, the transcription factor known as 'ELF5' inhibits sensitivity to estrogen very early in the life of a breast cancer cell.

In 2008, Associate Professor Chris Ormandy from Sydney's Garvan Institute of Medical Research showed that ELF5 was responsible for the development of the estrogen-receptor-negative cells in the breast during pregnancy that produce milk.

In the current study Ormandy, in collaboration with Drs Maria Kalyga and David Gallego-Ortega, has shown that the same molecular decision occurs in breast cancer and that ELF5 has the ability to change an existing tumour to an estrogen-insensitive type.

The team has also described the genetic mechanisms by which ELF5 opposes the action of estrogen, and has shown that it is possible to alter the subtype of breast cancer by manipulating ELF5 levels. The findings are published in the online journal PLoS Biology.

"This work tells us that cancers which become refractory to anti-estrogen treatment often do so by elevating their levels of ELF5 and becoming functionally estrogen receptor negative," said Ormandy.

"This raises the therapeutic option of manipulating ELF5 levels to treat breast cancer."

"As ELF5 is intracellular, this could possibly be done with small molecule therapies that target protein-to-protein interactions, or with small inhibitory RNAs."

"There is also the possibility of testing ELF5 levels in tumours to predict response to treatment and therefore guide treatment decisions."

"Our key discovery here is that by simply manipulating one transcription factor we can change the subtype of breast cancer."

Intel Patents Gaming System With "Safety Features"

Game systems that rely on gesture recognition for data input carry a potential for a player to unintentionally collide with surrounding objects.

We have grown accustomed to such scenarios since Nintendo decide to attach wristbands to its Wii Remotes and overeager Kinect players.

Intel has an idea to prevent objects from being damaged and players from being hurt. The company was just granted a patent that envisions proximity sensors to be built into game consoles that would trigger an alert or a game to be interrupted when a player may be too close to an object. From the patent:

"The gaming console may use the images of the full body movement and the images of the surrounding objects to determine if the user is close to the surrounding objects, which may cause collision with the surrounding objects. If the user is close to the surrounding objects, the gaming console may generate alert signals that may avoid or minimize the probability of collision of the user with the surrounding objects."

The patent was filed somewhat late, in March 2011, but was granted quickly in December 2012. The patent consists of hardware and software claims, where the hardware covers:

- a first set of sensors [that] continuously capture the first images of the body movement of the player
- a first set of sensors [includes] a camera to capture the first images of the body movement of the player
- a first set of sensors [that includes] a depth sensor to capture depth value of the first images of the body movement of the player
- a second set of sensors [that captures] the second images of the objects surrounding the player at regular intervals of time
- a first processor to determine proximity values based on a plurality of first position values and a plurality of second position values
- a first processor [to] generate an alert signal and pause signal if the proximity values are within the threshold values
- a second processor coupled to the first processor, wherein the second processor is to pause the game in response to receiving the pause signal

It would be unreasonable to conclude that Intel is suddenly interested in the video game console business. However, if we remember that Intel's core business intent is to sell more micro chips every year, then such patents make complete sense for the company.

Children, many ill, would be victims of Russia ban on U.S. adoption

MOSCOW (Reuters) - Family Christmas cards and smiling snapshots of children sent by their adoptive American parents fill Galina Sigayeva's office in Russia's second city St Petersburg.

Many of them were crippled by illness and in desperate need of medical care before her agency helped organise their adoption into U.S. families, she recalls.

Children's rights campaigners say children like these will suffer most if President Vladimir Putin approves a law barring American adoptions that has been rubber-stamped by Russian lawmakers. The act retaliates against a new U.S. law that will punish Russians accused of human rights violations.

Critics of the bill say Russian orphanages are woefully overcrowded and the fate of vulnerable children should not be used as a bargaining chip in a bilateral feud.

"These children are not even offered to foreigners until they get a certain number of (adoption) refusals from Russians," said Sigayeva, a neatly styled brunette who heads the New Hope Christian Services Adoption Agency.

"These are children with complicated diagnoses, really complicated. They are very ill children."

She smiled as she flipped through photos of children embraced by their adoptive parents, playing with family pets and enjoying presents and other trappings of holiday cheer.

"What surprises me is that here they all look so healthy, so fantastic, but you should see what they look like when they are taken from here," Sigayeva said.

"Some had to be carried to the border. We had a girl with hepatitis whom we helped from the emergency room."

Both sides in the heated debate surrounding the bill agree Russia's orphanage system is overwhelmed, riddled with corruption and most failing to place children in families.

More than 650,000 children are considered orphans in Russia - though some were rejected by their parents or taken from dysfunctional homes. Of that total, 110,000 lived in state institutions in 2011, according to the Ministry of Science and Education.

By contrast, in the United States - which has more than twice Russia's population - about 58,280 children were living in group homes and institutions last year, according to the U.S. Department of Health and Human Services.

Adoptions by Russian families remain modest, with some 7,400 adoptions in 2011 compared with 3,400 adoptions of Russian children by families abroad.

Russian politicians say it is an embarrassment that the country cannot care for its own, and supporters of the measure argue it will help stimulate reform and domestic adoptions.

"Foreign adoption is a result of the state and society's lack of attention to orphans ... It is, if you will, a result of our indifference," Prime Minister Dmitry Medvedev told officials at a ruling party congress last week.

American families adopt more Russian children - 956 last year - than those of any other country. Of the children adopted by Americans in 2011, 9 percent - or 89 - were disabled, according to official Russian figures.

Opponents of the legislation, who include senior officials such as Foreign Minister Sergei Lavrov, say politicking should not deprive orphans of this chance at better life.

"Russia is not able to provide for all its orphans," Boris Altshuler, director of the Moscow-based Rights of the Child advocacy group, said. "Although 1,000 is a small fraction - it was a help."

Deputy Prime Minister Olga Golodets has said the ban would violate international treaties on child rights, and the Kremlin's own human rights council called it unconstitutional.

"AMERICAN ROULETTE"

The ban responds to a U.S. law known as the Magnitsky Act, which punishes Russians suspected of being involved in the death in custody of anti-graft lawyer Sergei Magnitsky in 2009, and of other human rights violations, by barring them from entering the United States.

In a pointed echo of the Magnitsky Act, Russia's legislation is named the Dima Yakovlev law, after a Russian-born toddler who died of heat stroke after his American adoptive father left him locked in a sweltering car.

His death and that of 19 other Russian-born children in the hands of U.S. citizens in the last decade has helped drive support for the bill and for tougher adoption rules in a deal with the United States in June.

"It's American roulette," said Pavel Astakhov, Russia's Children's Rights Commissioner and a supporter of the ban.

"One handicapped girl from Russia got lucky. She was Jessica Long - a Paralympic champion. Another did not. She was Masha Allen ... who was raped by her paedophile adoptive father."

DISABLED CHILDREN

If Putin signs it into law, the ban will come into force on January 1, most immediately affecting the fate of children whose adoption is in the works.

The placement of 46 children with American families will be cancelled, Astakhov told the Interfax news agency on Wednesday.

"There is terrible irony in the fact that America's decision to speak out against human rights violations may cause the Russian government to deny many thousands of Russian orphans the possibility to grow up in loving, adoptive families," said Chuck Johnson, president of National Council For Adoption, a non-profit advocacy group based in Alexandria, Virginia.

Sigayeva, of the New Hope Christian Services Adoption Agency, said a six-month halt on American adoptions until a new bilateral deal entered force in November showed how it would aggravate problems in Russia's strained child-protection system.

"Hospitals were overwhelmed. There was no room in orphanages or hospitals for children whom their parents had rejected. So what's next then?" said Sigayeva, whose agency has helped place some 200 children in American families since 1992.

Advocates who work with disabled children say a reform proposal drafted by Astakhov ignores their plight. They say it calls for a reduction in the number of institutions caring for children with disabilities without explaining how they will find foster homes and medical care.

"No concrete measures are being suggested. Nothing exists but a lot of children's pain, which will only increase now," said Sergei Koloskov, a campaigner for children with Down's Syndrome.

"They are being left parentless in addition to being ill."

CAR rebels say they don't intend to seize capital

BANGUI, Central African Republic (AP) ? A leader for the rebels seizing towns across Central African Republic said Wednesday that his fighters do not intend to advance on the capital and remain open to dialogue with the government.

The announcement comes as fear grows in Bangui, a city of about 600,000 people, that government forces could face off against rebels here.

While the rebels have taken hold of at least 10 towns, the closest one is still 385 kilometers (240 miles) away.

Rebel Col. Djouma Narkoyo said Wednesday that his forces have continued taking towns in recent days because government forces are attacking their positions.

"Our intention is not to take Bangui," he said, when reached by phone. "We still remain open to dialogue."

There is unease, though, in the capital of Bangui. Residents here say they're skeptical about the claim the rebels don't intend to try and seize this city.

"We are afraid by what we see happening in our country right now," said Leon Modomale, a civil servant in the capital. "It's as if the rebels are going to arrive in Bangui any moment now because there are too many contradictions in their language."

Protesters gathered outside the French and U.S. embassies in Bangui on Wednesday, calling on the countries to help halt the rebel advance.

The rebel advance began earlier this month, with a push by the Union for the Democratic Forces for Unity, known by its French acronym of UFDR in this former French colony.

The group signed an April 13, 2007 peace accord, which paved the way for the fighters to join the regular army, an accord that the group's leaders say was never properly implemented.

Central African Republic is a desperately poor, landlocked country that has suffered numerous rebellions since independence from France.

Despite the nation's wealth of gold, diamonds, timber and uranium, the government remains perpetually cash-strapped.

U.S. special forces troops have deployed to Central African Republic among other countries in the region in the hunt for Joseph Kony, the fugitive rebel leader of the notorious Lord's Resistance Army.

WASHINGTON, DC December 26, 2012 -- Scientists have sequenced the genome of the fungus Pneumocystis jirovecii, an advancement that could help identify new targets for drugs to treat and prevent Pneumocystis pneumonia, a common and often deadly infection in immunocompromised patients. The study will be published on December 26, 2012 in mBio, the online open-access journal of the American Society for Microbiology. The organism cannot yet be isolated and grown for study in the laboratory, so details about Pneumocystis pneumonia, the biology of P. jirovecii, and its pathogenicity are hard to come by. The genome sequence represents a wealth of new information for doctors and researchers tackling this disease.

Pneumocystis pneumonia is an opportunistic infection that strikes most often in individuals with diminished immune systems. The corresponding author of the study in mBio, Philippe Hauser of the Centre Hospitalier Universitaire Vaudois and University of Lausanne, in Switzerland, says the disease gained importance in the 1980s.

"Recognized first among malnourished infants, P. jirovecii pneumonia became a public issue with the advent of the HIV epidemic," says Hauser. Today, the disease most commonly affects HIV-infected persons who are unaware of their status as well as solid organ transplant recipients and patients with hemato-oncologic or autoimmune diseases. Since the organism cannot be grown in the lab for study, researchers have long made do with studying P. jirovecii's lab-friendly relatives, species that infect animals and plants, in order to explore the secrets of the human disease.

"It is obviously better to study [P. jirovecii's] genes rather that those of Pneumocystis species from animal models. The genome has both medical and evolutionary interests for the scientific community," says Hauser.

Under normal circumstances, scientists sequencing the genome of a microorganism simply extract DNA from thick cultures of cells they grow in the lab. Since they were unable to grow P. jirovecii cells for their genomic DNA, Hauser and his colleagues took a different approach. They took a sample of bronchoalveolar lavage fluid from an individual infected with Pneumocystis pneumonia, then concentrated the P. jirovecii cells using immuno-precipitation and created copies of the DNA in the sample using a technique called random DNA amplification. This mixture of DNA strands, from P. jirovecii, human, and other microbes from the lungs of the infected patient, was then sequenced using high throughput technologies.

According to Hauser and his colleagues, the fact that the sequence data represented DNA from many different species created the biggest challenge they faced. "The major challenge of the study was the in silico sorting of the reads out of a mixture representing the human host and different organisms present in the lung microbiome," he says. This challenge was met through a collaboration with Marco Pagni of the Vital-IT group of the SIB Swiss Institute of Bioinformatics, who provided indispensable expertise and infrastructure.

Once the sorting task was accomplished, the researchers assembled the sequences into a genome and attempted to identify the functions of P. jirovecii's genes. This is the first time scientists have assembled the genome of a fungus from a mixed pool of DNA from a single source, often called a metagenome. Their analyses reveal a surprising fact: P. jirovecii is a parasite that must live within the human body to survive.

P. jirovecii lacks the genes necessary for creating some of the essential ingredients of life, a hallmark of obligate parasites, organisms that must rely on another creature for sustenance. "It implies that they need their host to provide these molecules. Thus, this has been quite an important finding which implied that human beings represent the reservoir of this pathogen," says Hauser. This is useful information, since it means that people are the only significant source of the organism and that both infected people and healthy carriers represent the only control points for limiting the spread of the disease.

The genome also shows that P. jirovecii apparently lacks the ability to make toxins and virulence factors, molecules that enable a microbe to invade and take advantage of its host. This makes sense, since P. jirovecii does not cause disease in healthy people, but only runs out of control when it is not confronted with an immune response.

In the study of infectious disease, access to the genome of a pathogen provides new information that can be pivotal in combating the diseases is causes. The hope is that the genome of P. jirovecii will lead to new advances in therapies for those suffering from Pneumocystis pneumonia. The current drugs of choice for treating Pneumocystis pneumonia are antifolates, but certain isolates of P. jirovecii have already developed resistance to antifolates, an ability that is very likely to spread. Now that the genome of P. jirovecii is assembled and available to researchers all over the world, scientists can tease out clues about the organism that will help identify targets for some badly needed new drugs.

PFT: Big decisions face Giants in offseason

TebowGetty Images

The soap opera continues, more than a week after the fat lady finished singing.

Quarterback-of-all-trades Tim Tebow has addressed with the media the report that he asked to be omitted from the team?s Wildcat package after being informed that he wouldn?t be the starting quarterback in Week 16.

?I never said, ?Hey, I don?t want to do anything.? I won?t do anything,?? Tebow said Wednesday, via Manish Mehta of the New York Daily News.? ?That wasn?t the talk at all.? He knows that.? And everybody on this team knows that I would never not to do something if I was asked.? That?s what?s disappointing.? People saying, ?Oh, you quit.? Or you didn?t do this.?? That was not it at all.? It was just me asking to get an opportunity to play the position I love, which is quarterback.? It wasn?t me asking out of anything.?

Tebow acknowledges that coach Rex Ryan may have misinterpreted the message, and Tebow says he wishes he would have been ?more clear? when meeting with Ryan.

?I was definitely disappointed and frustrated and I let him know that,? Tebow said.? ?Just asked for an opportunity to play quarterback.? [On Friday, I told him that] I would do anything for this team like I have all year from punt to hands team to catching passes . . . whatever I could do. He appreciated that. He understood. We got a great relationship still.?

And that?s where the logic falls apart.? Tebow wouldn?t have had to tell Ryan that Tebow would do whatever the team needed on Friday.? Ryan would have already known it.

Tebow said he was bothered by the notion that he?s not a good teammate, which was perpetuated by whoever from the Jets organization told multiple reporters that Tebow asked to be left out of the Wildcat package.

?You work your whole life to build a reputation,? Tebow said.

That line, of course, will do nothing to convince those who think that Tebow is concerned more about his image than anything else.? Indeed, it will make the skeptics even more suspicious that there?s a disconnect between Tebow and his ?reputation.?

For that reason, it would make more sense for Tebow to admit that, yes, he?s human.? And, yes, he sometimes has bad days.? And, yes, he was frustrated last week.? And, yes, he made it known that if he?s not the starter he doesn?t want to be the gimmick, either.

Saying that would go a much longer way toward helping his ?reputation? than the clumsy manner in which he has handled the current situation.

Astronauts Celebrate Christmas on Space Station

Christmas in orbit might not look exactly like the holidays on Earth, but the astronauts living on the International Space Station this holiday season try to make the orbiting science laboratory as homey as possible.

The six members of the station's Expedition 34 crew, three of whom just arrived last week, will all be spending Christmas and New Years Day aboard the spacecraft, but that doesn't mean they don't get to celebrate. Hundreds of miles above the Earth's surface, the spaceflyers will eat, exchange gifts, and be merry during Christmas and when welcoming in the New Year. ?

The space station crew will be off duty for both Christmas Eve and Christmas. That means that they won't need to work on any of the 110 experiments aboard the station, and they can take as much time for meals as they want, NASA spokeswoman Nicole Cloutier-Lamasters told SPACE.com.

The space station residents have a few different decorating options available to them. An earlier expedition left the crewmembers a Christmas tree and stockings made from nomex, a flame resistant fiber that's safe to stow onboard.? [Holidays in Space: Astronaut Photo Album]

The week before Christmas was an eventful one for the orbital crew as well. Canadian Space Agency astronaut Chris Hadfield, NASA astronaut Tom Marshburn, and Russian cosmonaut Roman Romanenko arrived on Friday (Dec. 21), joining Kevin Ford of NASA, and cosmonauts Oleg Novitskiy and Evgeny Tarelkin to fill out the $100 billion science laboratory to its usual 6-person capacity.

The spaceflyers also have some presents to look forward to. The Progress 48 cargo freighter ? a robotic Russian supply ship that launched in early August of this year? carried more than just basic supplies to the ISS. The Progress also brought holiday presents for the spaceflyers who'd be spending Yuletide in space.

As well as a traditional meal complete with turkey and candied yams, the crewmembers will also get the chance to video conference with their families. This is a particular treat because video chatting is usually possible only once a week, and involves a lot of planning for mission control and the spaceflyers.

Despite NASA's best efforts to make the holidays in space as warm as they are on Earth, that doesn't mean astronauts won't get homesick.

Marshburn, for example, has a 10-year-old daughter.

"That'll be tough, thinking about her waking up in the morning, enjoying things," said Marshburn during a preflight interview with NASA, "but the fact is we've got some technology that'll allow me, hopefully through an internet or I guess an internet protocol session, to be able to join in with them and see their faces and they can see me. It'll be a little tough for me, as it would be for anybody, but I think the price is certainly well worth it, to be up there."

Hadfield celebrated the holidays with his family before leaving for this, his second visit to the space station. Most of his family is spread around the world, Hadfield said during another preflight interview with NASA, so they celebrated the season early by flying to Kazakhstan to spend time with each other before his liftoff from the Baikonur Cosmodrome there. Hadfield is also an avid musician and guitar player, so music ? and a guitar that lives on the station ? will likely be a part of the Christmas celebration, Cloutier-Lamasters said.?

For his part, Romanenko said the prospect of a space Christmas is exciting.

"I think it will be a big adventure, a big moment in our space life," Romanenko said in a NASA preflight interview. "And we'll be dressing up, we'll be decorating the station, we'll put up a Christmas tree, maybe we'll have some presents that will arrive on the cargo vehicles, which of course will make us very happy and will support us during this evening, this special time."
